| Opis: | Usługi badawcze usługa: Stimulation and fixation of murine blood Opis Murine blood stimulation and fixation will be performed based on the exemplary protocol: 1. Collect blood samples into heparin-coated vials meeting the following specification: Sodium heparin blood collection vials o for small volume blood collections from 250-500 ul; o with push cap; o dimensions: 8 mm x 43mm; 2. Transfer 20ul of blood/animal into single well of PP 96-well V-bottom plate (or Eppendorf-like tubes). 3. Add 5ul/well (or Eppendorf-like tube) of 5x conc. anti-CD3 (25 ug/ml, final anti-CD3 concentration is 5 ug/ml) in plain RPMI. Incubate 60min, 37oC at 5%CO2. If using Eppendorf-like tubes, they should not be tightly closed during incubation. 4. During stimulation of samples prepare assay buffer as follows: Place the 1x Lyse/Fix buffer to water bath at 37oC for 25 minutes. 5. Fix samples immediately with warm 1x Lyse/Fix buffer to stop signaling cascades (add 180ul Lyse/Fix/well to all samples). 6. Incubate the cells at 37°C for 10 to 12 minutes. 7. Freeze and store the samples at -80oC and ship to Ryvu Therapeutics S.A. Lyse/fix buffer must meet the following minimal specification: o for use with intracellular flow cytometry; o reported to preserve the light scattering properties of the cell and may be used on human and mouse whole blood; o Storage Buffer: Aqueous buffered solution containing formaldehyde and proprietary ingredients; o can be used for lysing red blood cells and for fixing leukocytes in one step; o can be stored at room temperature; Antibodies to be used in the protocol must meet the following minimal specification: Purified anti-mouse CD3? monoclonal antibody o Isotype: Armenian Hamster IgG; o Clone: 145-2C11; o formulation: 0.2 um filtered in phosphate-buffered solution, pH 7.2, containing no preservative; o endotoxin level is <0.01 EU/ug of the protein (<0.001 ng/ug of the protein) as determined by the LAL test; o concentration: between 2 mg/ml and 3 mg/l; o antibody was purified by affinity chromatography; Purified Armenian Hamster IgG Isotype control antibody o Isotype: Armenian Hamster IgG; o Clone: HTK888; o formulation: 0.2 um filtered in phosphate-buffered solution, pH 7.2, containing no preservative; o endotoxin level is <0.01 EU/ug of the protein (<0.001 ng/ug of the protein) as determined by the LAL test; o concentration: between 2 mg/ml and 3 mg/l; o antibody was purified by affinity chromatography; The material (blood from mice) will be provided by the Sponsor. Contractor should provide all the reagents and necessary equipment to perform the service according to the protocol. Contractor should declare a possibility to process up to 1000 blood samples within 14 months according to the protocol listed above. It is assumed that there may be up to 10 shipments of the processed (stimulated and fixed) blood samples from Contractor to Sponsor within 14 months.
